BAX is required for induction of apoptosis in LY-treated LNCaP cells. LNCaP were transfected with control siRNA, BAD siRNA, BAX siRNA or both BAD siRNA and BAX siRNA. At 48 h post-transfection, cells were serum starved followed by treatment either 10% serum-containing medium (serum) or LY in serum-free medium (LY). (a) Western blot of lysates collected 72 h post-transfection with anti-BAD and anti-BAX antibodies demonstrated specific silencing of BAD and BAX. β-Actin was used to show equal loading. Apoptosis in LY-treated cells at 6, 12 and 24 h post-treatment were shown by (b) % of cells in sub-G1 phase and (c) caspase 3 activity.
