Activated M-Ras supported the factor-independent colony formation and differentiation to macrophages and neutrophils. Bone-marrow cells transduced with retroviruses expressing GFP alone or constitutively active M-Ras or H-Ras were selected for by culture for 48 h in the presence of puromycin as described and washed free of growth factors. (a) 5 × 103 cells were seeded in triplicate into medium gelified with methylcellulose and with or without a mixture of synthetic IL-3 (2 μg/ml) and IL-6, SCF and GM-CSF at 10 ng/ml, and numbers of colonies were counted at day 7. Values are the mean±s.d. of the triplicates, and data are the representative of three independent experiments. (b) 2 × 103 cells were plated in plasma clots in the absence of growth factors as described above. Plasma clots were dried and stained with Hema stain at day 7. Immature neutrophils are recognizable by their ring-shaped nuclei
