The effects of inhibition of the activities of PI3K, ERK and p38 MAP kinase on the proliferation of bone-marrow cells expressing activated M-Ras or H-Ras. Bone-marrow cells transduced with retroviruses expressing GFP alone or constitutively active M-Ras or H-Ras were selected for 2 days in the presence of puromycin as described and washed free of puromycin and growth factors. (a) Puromycin-selected cells (1 × 105/ml) were seeded into the wells of 24-well plates in the presence or absence of PD98059 (25 μ M) or Ly 294002 (25 μ M). The cells were counted 5 days later; the numbers are presented as mean±s.d. (b) Prior to the culture, washed puromycin-selected cells were also incubated without serum for 3 h, lysed and the cell lysates subjected to SDS–PAGE and immunoblotting blotted with anti-phospho-Erk antibody and anti-β-actin antibody. (c) Puromycin-selected cells (1 × 105/ml) were seeded into the wells of 24-well plates in the presence or absence of SB 203580 (5 μ M). The cells were counted 5 days later; the numbers are presented as mean±s.d.
