SNIP1 induction following serum stimulation. (a) Analysis of serum-induced proteins in U-2 OS cells. U-2 OS cells were serum starved for 96 h and then released back into the cell cycle by readdition of serum. Whole-cell lysates were harvested at the indicated times post serum addition, resolved by SDS–PAGE and then immunoblotted using anti-SNIP1, cyclin D1, c-Jun and β-actin antibodies. (b) Serum stimulation induces SNIP1 expression. U-2 OS cells were serum starved as outlined in (a). Cells were permeabilized and stained with the 2B12 SNIP1 monoclonal antibody. DAPI stain was used to stain the cellular DNA. Immunofluorescence analysis was performed as in Figure 4. (c) c-Jun induces SNIP1 expression. U-2 OS cells were transfected with indicated levels of RSV c-jun expression plasmid. RSV controls were included as appropriate such that all transfections had equivalent levels of expression plasmid. Whole-cell lysates were harvested 48 h after transfection, resolved by SDS–PAGE and then immunoblotted using anti-SNIP1, c-Jun and β-actin antibodies. ns indicates the position of a nonspecific band seen in some experiments
