SNIP1 depletion does not lead to activation of p53. (a, b) p53 protein and mRNA levels do not increase following SNIP1 knockdown in U-2 OS cells. U-2 OS cells were transfected with the indicated SNIP1 and control siRNA oligonucleotides. At 48 h after transfection, whole-cell protein lysates or RNA were prepared, and either resolved by SDS–PAGE and immunoblotted with anti-SNIP1, anti-p53 or anti-β-actin antibodies or subjected to semiquantitative PCR analysis for SNIP1, p53, Hdm2 and GAPDH expression. (c) U-2 OS cells were transfected with 1.5 μg of p21 luciferase reporter plasmid and either 3 μg of a control siRNA expression plasmid or 1 μg each of three SNIP1 siRNA plasmids (3 μg total). Cells were harvested 48 h after transfection and the luciferase activity was measured. (d) U-2 OS cells were transfected with the indicated SNIP1 and control siRNA oligonucleotides. At 48 h after transfection, whole-cell lysates were prepared, immunoblotted with anti-SNIP1, anti-p21 and anti-β-actin antibodies
