Tuberin expression does not activate Rac1 and cdc42. Cells were serum-starved 16 h before the assay. GTP-bound Rac1 and cdc42 was pulled down from 750 μg protein lysate using GST-Pak1-p21 binding domain agarose beads, and immunoblotted using anti-Rac1 and anti-cdc42. Whole cell lysates were used for assaying total Rac1 and cdc42. Results are representative of three independent experiments. (a) MDCK cells. (b) ELT3 cells
