Methylation profile of the Sp1 mutation, promoter deletion and ATAAA deletion constructs transfected into LNCAP. (A) The promoter deletion construct and (B) The Sp1 mutation construct were stably transfected into LNCAP cells and DNA harvested after 22, 45 and 70 doublings (22d, 45d and 70d). (C) The ATAAA deletion construct was stably transfected into LNCaP cells and DNA was isolated after 22 doublings (22d). The methylation status of the CpG island from CpG site −41 to +10 was analysed by bisulphite sequencing a number of clones from 2–3 PCR reactions. Clones from the promoter deletion construct indicate that CpG sites −2 to −8 are deleted. Clones from the Sp1 mutation construct show grey shaded circles at the mutated Sp1 CpG sites that are not available for methylation
