Recreation of IL-21R/BCL6 fusion gene and expression of Bcl-6 in COS-7 cells. (a) Schematic presentation of BCL6-expression plasmids that are driven by the SV40, BCL6 and IL-21R promoters (the sequences of the latter two are presented in Figure 5a). Restriction sites: H, HindIII; X, XbaI; G, BglII. The XbaI site within exon 4 is demethylated in DM1 (GIBCO/BRL). MCS, multiple cloning site. (b) Western blot analysis of COS-7 transfectants for Bcl-6 expression. FL-218 was to indicate the position of normal Bcl-6 protein. Cell lysates were prepared from 1×106 cells and an aliquot was loaded. (c) Indirect immunofluorescence of COS-7 cells transfected with BCL6-expression plasmids. The cells were sequentially labeled with a primary rabbit antibody against Bcl-6 and secondary anti-rabbit antibody labeled with Alexa 488.
