Figure 7

Transformation by a LMP1 deletion mutant. (a) Total RNA (5 μg) from MDCK cells (lane: MDCK), MDCK cells transfected with control plasmid (lane: neo), expression plasmid for LMP1 (lane: LMP1), LMP1 deletion mutants lacking carboxy-terminal a.a. 238–386 region and a.a. 187–351 region (lanes: 1–231 and Δ187–351, respectively) were electrophoresed in a gel, stained with ethidium bromide (bottom panel), and then examined by Northern blotting probed with 32P-labeled LMP1 and uPA cDNA fragments (upper and middle panels, respectively). (b) Left panels: subconfluent cultures of MDCK cells transfected with control plasmid and expression plasmid for LMP1 deletion mutants lacking carboxy-terminal a.a. 232–386 region and a.a. 187–351 region (panels: 1–231 and Δ187–351, respectively) are shown. Right panels: confluent monolayer of MDCK cells transfected with control plasmid and expression plasmid for LMP1 deletion mutants lacking carboxy-terminal a.a. 232–386 region and a.a. 187–351 region (panels: 1–231 and Δ187–351, respectively) were scraped by a plastic pipet, and wound-induced migration of cells was observed after 5 h.
