Is elevated cardiovascular risk a class effect of cyclo-oxygenase 2 inhibitors?

In 2004, an increased risk of cardiovascular events was observed in a randomized controlled trial of rofecoxib for polyp prevention, leading to discontinuation of that trial, withdrawal of rofecoxib from the market, and a focused review of other agents in this class.1 Within a few months, data showed that celecoxib and valdecoxib were also associated with increased cardiovascular risk.2, 3 Ultimately, these data and a comprehensive FDA review led to a black-box warning for celecoxib, and valdecoxib's withdrawal.
