T-cell receptor

T-cell receptors (TCRs) recognize small protein fragments (peptides) that are displayed on the surface of antigen-presenting cells. Each T cell expresses a unique TCR that is generated by randomly assorting genes. This ensures that T cells can respond (at the population level) to almost any infection.

Mucosal-associated invariant T (MAIT) cells express invariant TRAV1/TRAJ33 TCR-α gene segments and detect antigens presented by MR1. Here the authors show that atypical, MR1-restricted MAIT populations that include both Trav1+ and Trav1- cells are found in both Traj33-deficient mice and human peripheral blood.

The percentage of cancer neoantigens that are spontaneously recognized by T cells is generally very low. This protocol describes how CD8+ T cells from healthy donors can be used for enhanced targeting of these neoantigens.

Supraphysiological T cell activation by chimeric antigen receptor (CAR) contributes to T cell exhaustion and adverse events in CAR T cell therapies. Here the authors engineer a synthetic antigen receptor that integrates into the endogenous TCR complex, preserving natural regulatory circuits and achieving improved performance in mouse tumor models.

Some immune cells undergo processes that pose unique challenges to the 3D organization of their genomes. These include antigen receptor rearrangement, clonal expansion and the contortion of their nuclei. Here, Allan and colleagues discuss the latest insights into these processes from a structural genomics perspective.

For T cells, functional antigen receptors are selected in the thymus from a pre-selection repertoire by interaction with self MHCs. Here the authors show that specific, non-germline coded features located in the complementarity determining region 3 of the pre-selection antigen receptors are essential for the selection of MHC-restricted repertoire.

Anti-human leukocyte antigen (HLA) antibodies are important mediators of alloresponses, but structural insights on antibody:HLA interaction are still lacking. Here the authors provide a 2.4 Å structure of antibody:HLA complex, and also analyse HLA features important for other HLA-interacting molecules, to enhance our understanding of alloimmunity.
