Sp1 requires Zn(ll) for sequence-specific binding to DNA.
Inactivation of Sp1 by treatment with EDTA and recovery of DNA binding activity by addition of Zn(ll). The footprinting reactions contained no Sp1 (lane l); untreated Sp1 (lane 2); Sp1 + EDTA (lane 3); and apoSp1 incubated with water (as a negative control; lane 4), ZnSO4 (lane 5), CoSO4 (lane 6), or NiSO4 (lane 7).
