FRET visualizes the Ran gradient
(A) shows the scheme of the FRET biosensor. An importin beta-binding domain (IBB) is flanked with YFP at the N terminus and CFP at the C terminus. This biosensor increases its FRET signal when released from importin-beta by RanGTP. (B) The FRET signal in a metaphase cell (top) and the Xenopus metaphase extract (bottom). See that similar RanGTP gradient surrounds the chromosomes both in mammalian cell and Xenopus extract.

How does Ran GTPase control both protein import during interphase and spindle attachment to the chromosomes during M-phase? It all has to do with its concentration gradient in the cell.
