MUC1 upregulates the phosphorylation of IGF-1R. (a) HEK-293A, HS578T and MDA-MB 231 cells were transfected with pcDNA3 or pcDNA3-MUC1 for 48h. Lysates were analyzed for phosphorylated IGF-1R (pIGF-1R) and total IGF-1R levels. (b) Cell lysates from MDA-MB-231 Re/MUC1 clones and the Re/EV clone were collected and submitted to western blotting for pIGF-1R and IGF-1R. (c) MDA-MB-231 Re/MUC1 and Re/EV clones were transfected with scrambled siRNA (Sc) and MUC1 siRNA (siMUC1). Cell lysates were then submitted to western blotting to measure pIGF-1R and IGF-1R levels. (d) Human breast cancer cells expressing endogenous MUC1 were transfected with scrambled siRNA or MUC1 siRNA. pIGF-1R, IGF-1R and VEGF levels were measured by western blotting. (e) MDA-MB-231 Re/MUC1 and Re/EV clones were transfected with scrambled siRNA (Sc) or IGF-1R siRNA (siIGF-1R). Cell lysates were recovered and submitted to western blotting to measure VEGF and pAKT levels. Actin was used as an internal control. (f) Western blotting of mouse spontaneous breast cancer cells expressing human MUC1. pIGF-1R, IGF-1R and VEGF levels were measured by specific antibodies.
