Dose- and time-dependent induction of DNA demethylation and reexpression of SNCG in A549 cells by CSE. A549 cells were exposed to the indicated concentrations of CSE for 3 days or to a concentration of 2.5% CSE for different days. The medium was changed every day with fresh medium and CSE. By the end of treatment, genomic DNA and total RNA were isolated from untreated and treated cells. (a) Dose-dependent effects of CSE on SNCG mRNA expression and CpG island demethylation in A549 cells. (b) Time-dependent effects of CSE on CpG island demethylation in A549 cells examined by MSP (left panel).
