The authors carried out a proteomic analysis of purified NGF–TRKA endosomes and identified over 250 endosome-associated proteins. Many of these are known regulators of the actin cytoskeleton, which led the authors to examine the effects of actin modifying drugs on endosome assembly and transport. They found that actin stabilization with jasplakinolide completely abolished endosome transport in cultured sympathetic neurons, indicating that actin depolymerization is required to initiate the retrograde transport.

The authors found that the actin-severing and endosome-associated protein cofilin is required for retrograde transport, as knocking down cofilin with small hairpin RNA impaired endosome transport. This effect could be rescued by exposing distal axons to the actin depolymerizing drug latrunculin A, confirming that cofilin-mediated actin disassembly is key to the retrograde transport of endosomes.
