A safety net for successful mitosis

During mitosis, the SAC proteins mitotic arrest deficient 1 (MAD1) and MAD2 are recruited to unattached kinetochores to promote the formation of the mitotic checkpoint complex (MCC). The MCC sequesters CDC20 to prevent activation of the APC/C (anaphase-promoting complex, also known as the cyclosome) and hence delay anaphase. During interphase, MAD1 and MAD2 are associated with nuclear pore complexes (NPCs), and a large proportion of MAD2 is already bound to CDC20. Given that NPCs have been implicated in regulatory functions other than nuclear transport, Rodriguez-Bravo et al. investigated the functions of NPC-tethered MAD1 during interphase.
