To identify genes that cooperate with p53 to control proliferation, the authors executed a genome-scale RNA interference (RNAi) synthetic interaction screen in wild-type and p53-null cells, and compared the growth response of the two cell populations. The authors hypothesized that factors which differentially inhibit the growth of p53-null cells when they are depleted collaborate with TP53 in wild-type cells to maintain homeostasis. One of the three genes identified, UNR-interacting protein (UNRIP; also known as STRAP), encodes a core component of the survival of motor neuron (SMN) complex and was chosen for further study.
