Heterozygous mutations in the genes encoding ribosomal proteins (RPs) are emerging as important determinants of human disease and tumorigenesis. But what links Rp gene dosage and disease? In zebrafish, heterozygous loss of one of 17 of the 28 Rp genes results in the development of malignant peripheral nerve sheath tumours (MPNSTs), and zebrafish expressing a loss-of-function p53 protein also develop MPNSTs.
