Differentiation of mouse embryonic stem cells to insulin-producing cellsInsa S Schroeder et al.
This protocol from the laboratory of Anna Wobus describes how to form embryoid bodies from mouse embryonic stem cells followed by the induction of progenitors to pancreatic cells. After approximately 33 days in culture the differentiated cells form islet-like clusters that respond to glucose with the release of insulin.

Differentiation of mouse embryonic stem cells to hepatocyte-like cells by co-culture with human liver nonparenchymal cell linesAlejandro Soto-Gutiérrez et al.
In this protocol mouse embryonic stem cells are directed to differentiate into hepatocyte-like cells. The resulting cells express the hepatocyte specific genes albumin and CYP7a1.

Production of chimeras by aggregation of embryonic stem cells with diploid or tetraploid mouse embryosGuy S Eakin & Anna-Katerina Hadjantonakis
Mouse chimeras are produced in this protocol using either diploid or tetraploid morula. Tetraploid cells are formed by the electrofusion of day 1.5 post coitum embryos whereas diploid cells are collected from 8–cell embryos. The cells are then aggregated with embryonic stem cells to form chimeras.

Evaluation of purified hematopoietic stem cells from umbilical cord bloodChristopher Y Park, Ravindra Majeti & Irving L Weissman
Human hematopoietic stem cells are isolated from umbilical cord blood in this protocol. Cells that are Lin-CD34+CD38-CD90 are transplanted into laboratory animals and long and short term engraftment is measured.

Adult mouse hematopoietic stem cells: purification and single-cell assaysHideo Ema et al.
This protocol isolates mouse hematopoietic stem cells based on their surface marker expression. CD34−/lowc-Kit+Sca-1+lineage marker− cells are isolated using flow assisted cell sorting and then used in single cell assays to measure their multilineage repopulating activity.
