One hundred microgram of nuclear extract from SH-SY5Y cells was separated in a 10% SDS-PAGE. The separated proteins were transferred to Hybond-C extra membrane, which was then incubated with either the (a) wild-type or the (b) mutated probe, and the signals were visualized by autoradiography. The sequences of the probes are shown below. The underlined sequences indicate the locations of the GC-boxes and the N1 target site. The bold-type sequences are the mutated motifs.
