In their recent paper, Kukar et al.1 studied the effects of a number of drugs on beta-amyloid (Abeta) production in wild-type mice and a mouse model of Abeta deposition. They conclude that there are therapeutic agents that may augment Abeta42 production and thereby increase the risk of Alzheimer disease in individuals on long-term therapy with such drugs.
