Almost a decade ago, thalidomide was shown to be clinically active in patients with relapsed and refractory multiple myeloma.1 Combinations of thalidomide with dexamethasone or chemotherapy have now been tested in patients with newly diagnosed myeloma and have been shown to improve response rates. Thalidomide therapy can, however, be associated with potentially disabling adverse events, such as peripheral neuropathy, which may limit its long-term use.
