An increase in the size of the amyloid beta-peptide (Abeta42 versus Abeta40) may be a key factor in the pathogenesis of Alzheimer's disease. By altering the activities of enzymes involved in the metabolism of cholesterol and sphingomyelin, an increase in the Abeta42:Abeta40 ratio may cause dysfunction and death of neurons.

Drosophila G-protein signalling: intricate roles for Ric-8? pp1047 - 1049

Receptor-independent G-protein signalling regulates both the orientation and position of the mitotic spindle during asymmetric cell division. Several new studies show that the targeting of G-protein subunits to the membrane requires Ric-8, pointing to possible novel roles for this protein in both receptor-dependent and -independent pathways.

How do cells achieve specific and efficient control over a wide spectrum of activities in the context of a limited protein repertoire? A possible answer comes from the recent demonstration that utilization of distinct enzymes by the same A-kinase anchoring protein (AKAP) depends on the AKAP-bound substrate, thus vastly increasing the diversity of AKAP-mediated protein regulation.

Guidance molecules ensure that nerve cells grow in the correct direction during brain development. However, very little is known about the signals transmitted by these molecules. It now seems that netrin-1 — the first guidance molecule discovered — achieves this, in part, through activation of PITPalpha, a protein involved in lipid transfer.
