a, TCF4 is present in c-Jun immunoprecipitations (IP). Pre-incubation with GST−c-Jun, but not GST alone, prevents binding. b, 293T cells were transfected with Myc-tagged c-Jun, c-Jun4A and haemagglutinin (HA)-TCF4, Myc-Tag immunoprecipitations were performed and TCF4 protein levels were analysed by western blot. c, HEK 293T cells were transfected with HA-TCF4 or HA-DeltaN-TCF4, c-Jun or beta-catenin immunoprecipitations were performed, and TCF4, c-Jun and beta-catenin protein levels were analysed by western blotting. An asterisk indicates an unspecific band. d, 293T cells were transfected with c-Jun and beta-catenin plasmids or a pSUPER vector expressing a short hairpin RNA (shRNA) specific for TCF4 (pS-siTCF4), c-Jun immunoprecipitations were performed, and TCF4, c-Jun and beta-catenin protein levels were analysed by western blotting. In all experiments JNK activity was stimulated by treatment of cells with 5 mM anisomycin 1−2 h before immunoprecipitation.
