Abstract
GLUCOCORTICOID stimulation of alkaline phosphatase (AP) activity in HeLa cells is believed to be mediated by steroid receptors1,2. When synchronised HeLa cells are exposed to glucocorticoids, an increase in AP activity is initiated during the synthesis phase of the cell cycle3. This ‘S-phase dependency’ suggests that the necessary components for cellular response to glucocorticoids are produced or altered during this time. Because the interaction of the steroid with its cytoplasmic receptor seems to be the first major intracellular event of the action of glucocorticoids, we have measured the glucocorticoid receptors present throughout the cell cycle in synchronised HeLa cells. We report here that although receptors are present throughout the cycle, the number per cell varies markedly within the cell cycle in both cytosols and whole cells.
