Holography without Fringes in the Electron Microscope

Abstract
I REPORT here the successful solution of the phase problem for a negatively stained catalase crystal using diffraction and image plane pictures produced in an electron microscope. The results show the complete wave function, phase as well as amplitude maps, in-the microscope image plane. The computational method employed in achieving the solution has been described by Gerchberg and Saxton1 who demonstrated the technique on general mathematical functions; the method was later successfully applied to mathematical models of magnetic thin films2. I have now applied the method to a real biological crystal in the electron microscope.
