Cyclic AMP Mediates the Effects of Adrenaline on Cardiac Purkinje Fibres

Abstract

MANY of the biochemical actions of catecholamines and other hormones are mediated by alterations in intracellular levels of adenosine 3′,5′-monophosphate (cyclic AMP)1. For example, adrenaline stimulates glycogenolysis in heart muscle by increasing intracellular cyclic AMP levels. It is not clear, however, whether any of the effects of adrenaline on the electrophysiology of the heart are similarly mediated by this cyclic nucleotide2, although recent experiments indicate that exogenous cyclic AMP can accelerate the rhythmic beating of heart cells in culture3. We have studied the currents which underlie the action potential plateau in cardiac Purkinje fibres4–6 as a means of exploring the possible role of the cyclic nucleotide in modifying individual ionic current components. Our results strongly suggest that cyclic AMP does mediate the action of adrenaline on the plateau.
