SH-SY5Y cells transfected with PKC
antisense cDNA show a reduced sensitivity to PMA stimulation of sAPP release compared with wild-type cells. Cells were incubated for 2 h with serum-free medium alone or increasing concentrations of PMA (10 nM, 100 nM, 1 
M). Proteins secreted into the conditioned media were collected and subjected to Western blot analysis for sAPP. The blots were quantified by densitometry as described in the 'Materials and methods' section. Data are expressed as a percentage of basal release and are representative of three to four independent experiments.
