JC-1 vital staining of rat cardiomyocytes exposed to oxidative stress. Non-pretreated and pepstatin-A-pretreated (100 mum) cells were exposed to 1.5 mum naphthazarin for various amounts of time and then stained with 5 mug/ml JC-1 for 10 minutes. Mitochondria are green (Deltapsim m < -100 mV) and orange-red (Deltapsim m > -140 mV) in control cells but only green in cells with a decreased Deltapsim. Cells containing green or red/green mitochondria were counted, and the illustrated values are means plusminus sd, n = 4 (500 cells/sample). Cells pretreated with pepstatin A and then subjected to naphthazarin differed significantly from cells exposed solely to naphthazarin, as calculated by the Mann-Whitney U test. * p less than or equal to 0.05.
