PKC-delta immunofluorescence. Mesangial cells growth-arrested on glass cover slips were cultured in normal (5.6 mM) glucose or high (30 mM) glucose for 48 hours, followed by 10 minutes of stimulation with endothelin-1 (ET-1; 100 nM). PKC-delta was detected in fixed, permeabilized cells by confocal scanning immunofluorescence microscopy using monoclonal anti-PKC–delta antibody and FITC-conjugated secondary antibody (magnification times240).
