Supplementary figure 7. PKA and p38 are not required for Ad2 internalization and delivery to the cytosol. (A) The kinetics of viral uptake into HeLa cells treated with the PKA inhibitor KT5720 or the p38-inhibitor SB203580 was determined using [35S]methionine-labeled Ad2 and a cell surface trypsinization assay that measured the emergence of a trypsin-resistant form of the major capsid protein hexon. The fraction of trypsin-resistant hexon was plotted as a function of infection time. There is no difference between the inhibitor-treated and the control cells. (B) Quantitative EM analyses of viral particles in PKI-myr-, SB- and no drug-treated cells. There are no differences in the amounts of virus particles at the plasma membrane, in intracellular vesicles and in the cytosol at 30 min post-infection (p >0.1)
