Different GFP–Bax mutants display opposite effects on cell viability. (A) L929 mouse fibroblasts, and (B) and (C) Bax-/- cells were transiently co-transfected with the pGL3 luciferase reporter construct (0.25 mug) and different GFP fusion constructs of either wild-type or mutated Bax (1 mug). The C3-EGFP expression vector was transfected as a control. After 40 h of incubation, cells were harvested and processed for luciferase assays. Results were quantitated with a scintillation counter and displayed as a percentage of the control. The error bars show a mean standard error determined from at least three independent measurements.
