Aiolos transcription factor controls cell death in T cells by regulating Bcl-2 expression and its cellular localization

Transfection of Aiolos induces Bcl-2 expression in IL-2-deprived cells. Cells were transiently transfected with or without Aiolos using the DEAE–dextran method and stimulated with or deprived of IL-2 for 24 h. After transfection, protein extracts from 6times105 cells were separated in SDS–PAGE, transferred to nitrocellulose and probed with anti-Bcl-2 antibody. IL-2- (5 ng/ml) or IL-4-stimulated (60 U/ml) cells were used as positive and negative Bcl-2 expression controls, respectively. Protein bands were developed using ECL. Hybridization with pan-Ras is shown as an internal protein loading control. Expression of the transiently transfected Aiolos was confirmed by direct comparison of Aiolos protein levels in transfected and mock transfected controls. Molecular markers are shown. Similar results were obtained in three independent experiments.
