Following pretreatment of HEK-293 cells for 10 min with vehicle (Control), 30 muM DHS or 15 muM DMS, [3H]sphingosine was added to the cells at t0 and basal formation of [3H]SPP was determined for either 30 s or 2 min. Values shown are meanplusminusSD of two independent experiments, each performed in triplicate, and corrected for t0 count rates of 39plusminus5 c.p.m./106 cells. Pretreatment with DHS or DMS did not alter the cellular uptake of [3H]sphingosine.
