Open-channel blockers such as memantine inhibit the NMDAR preferentially when it is excessively (pathologically) activated but have little if any effect on normal synaptic transmission. (Glu=glutamate; Gly=glycine; NR1 and NR2 are subunits of the NMDAR that is probably composed of a tetramer of subunits)
