Effect of JAK inhibition on ERK1/2 phosphorylation (a) and effect of MEK/ERK1/2 inhibition on STAT-3 phosphorylation (b). Cells were preincubated for 48 h in serum-free medium and hypoxia and pretreated with either 10 mu M U0126 or 50 mu M AG490 for 60 min, and then were treated with 20 ng ml-1 PlGF for 15 min. Cell lysates were run on 10% SDS–PAGE gels and then transferred to a polyvinylidene difluoride (PVDF) membrane and treated with the anti-phospho-ERK1/2. The membranes were stripped and reblotted with anti-phospho-STAT-3 antibody. Results are expressed as OD ratio using actin as control.
