Hiding from the inflammasome

the T3SS can activate the inflammasome, but this activation is inhibited by the secreted effector YopK

To investigate the effect of the T3SS during infection by pathogenic yersiniae, the authors generated mutant Yersinia pseudotuberculosis that expressed the T3SS but not the translocated virulence proteins (T3SS bacteria). These mutants triggered rapid caspase 1 activation and cell death in infected macrophages; by contrast, these responses were either absent or triggered 4 hours later following infection with YopJ-deficient or wild-type bacteria, respectively. As inflammasome activation is necessary for caspase 1 activation, this suggests that the T3SS itself can activate the inflammasome and that a translocated protein distinct from YopJ might inhibit this activation.
