By using biochemical fractionation, the researchers found that mammalian WNT3A or the fly homologue WG (Wingless) and the exosomal marker TSG101 co-purified with vesicular particles from cell culture supernatants. The purified exosomal fraction from WNT3A-transfected cells induced WNT activity, whereas depletion of exosomal WNTs by ultracentrifugation reduced this activity, thus demonstrating the signalling capability of exosomal WNTs.
