Gene dosage in mice—BAC to the future

Despite remarkable strides in the human genome project, the task of understanding the function of each gene remains formidable. So, there is an urgent need for methodologies that illuminate gene function in mammals. One such tool is described on page 327 of this issue by Xiangdong Yang and colleagues1. They use bacterial artificial chromosomes (BACs) to 'dose' Zipro1, which encodes a zinc-finger transcription factor (known as RU49), in mice. In so doing, they discover that Zipro1 regulates cell growth—increasing dosage resulted in a sixfold elevation in expression, which, in turn, enhanced cell proliferation in the cerebellum and skin. In contrast, no distinctive phenotype was obtained by a loss-of-function approach.
