Verification and initial annotation of the NIA mouse 15K cDNA clone set

Expression profiling of thousands of genes on cDNA microarrays is now a powerful tool in biology and medicine. To facilitate such work in mouse models, we assembled a set of 15,247 unique oligo(dT)-primed cDNA clones (“NIA Mouse 15K”) based on 52,374 3′-expressed sequence tags (3′-ESTs) that are primarily derived from early embryonic cDNA libraries1. The clone set has been distributed freely and is in use at more than 100 sites2 (for access information, see http://lgsun.grc.nia.nih.gov/cDNA/cDNA.html).

To verify clone identity and obtain protein-coding information, we re-sequenced the cDNA clones, with an average insert size of 1.5 kb, from both 3′ and 5′ ends (GenBank BG062929–BG088954). We compared sequence tracts, averaging 516 bp with less than 10−2 errors per base pair3, with the 3′ ends of the source (parent) clones using BLAST v. 2.0.14 (ref. 4). We thus identified and corrected errors committed in handling the clones.
