Abstract
THE major chemical constituent of the lampbrush chromosomes and nucleoli of newt oocytes is known to be protein1,2. We report here the semi-bulk fractionation of germinal vesicles, isolated manually from oocytes of Triturus cristatus carnifex, into nucleoplasm and a “chromatin” preparation consisting of lampbrush chromosomes, nucleoli, nucleoplasmic granules and nuclear membranes. Two properties of the proteins in the chromatin have emerged. First, protein–disulphide-bond crosslinks make a definite contribution to the morphological integrity of the isolated lampbrush chromosomes and nucleoli. Second, the non-histone proteins of oocyte chromatin consist predominantly of two major polypeptide species.
