Presentation of an Antigenic Determinant from Hen Egg-White Lysozyme on the Flagellar Filament of Escherichia Coli

Abstract

We attempted to take an antigenic determinant of hen egg-white lysozyme and present it on the Escherichia coli flagellar filament by inserting a lysozyme-derived fragment into the central region of the filament component protein, flagellin. Random digests of the lysozyme gene were inserted into the central region of the flagellin gene, and a motile strain whose motility was inhibited by anti-lysozyme serum was selected. It had a chimeric flagellin containing an 11-residue lysozyme-derived sequence (Trp63-Arg73) in the central region. In an enzyme-immunoassay, flagellar filament composed of the chimeric flagellin reacted well with anti-lysozyme antibody. However, this filament had no ability to induce antibodies in vivo that would bind to intact lysozyme indicating that the lysozyme fragment on a flagellar filament exhibits immunological properties similar, but not identical, to those of intact lysozyme. They also prove that the flagellin region into which the foreign fragment has been inserted is exposed on the surface of the filament. The chimeric flagellin system may be useful for analysis of epitopes of a protein.
